Intratracheal Instillation in Rats to Assess Nanomaterial Toxicity


Although nanomaterials have immense potential applications, some respirable types have toxic characteristics that can be detrimental to human health. To prevent such health hazards, determining their pulmonary toxicity using inhalation exposure studies is necessary. However, performing inhalation exposure studies for each type of nanomaterial is costly and impractical. We therefore propose using intratracheal instillation in rats as a simple and inexpensive method to identify toxic nanomaterials that require further inhalation studies.


In our inter-laboratory study, we investigated whether intratracheal instillation was a useful screening method to detect the pulmonary toxicity of nanomaterials. Five independent research institutions followed a standardized procedure to perform intratracheal instillation of nickel-nanoparticles (NiO-NP) in rats. Eight-week-old rats were exposed to NiO-NP by a single intratracheal instillation at doses of 0 (vehicle control), 0.2, 0.67, and 2 mg/kg per body weight. On the third day after instillation, histopathological analysis of the lungs was conducted. Any observed alterations of the lung tissue were classified into grades, depending on the severity of the lesions.


Our findings show that the severity of pulmonary lesions, such as inflammation, degeneration, and necrosis correspond to increasing doses of NiO-NP. Standardizing intratracheal instillation procedures across laboratories enabled comparisons of the histopathological results, and similar toxicity trends were detected at all five research institutes.


Thus, our analysis confirms that intratracheal instillation in rats is an effective method that can be used to assess the pulmonary toxicity of nanoparticles and to help prevent human exposure to hazardous nanomaterials.
